📖 Instruction for Use

Dosage

• Standard Adult Dosage: 1.125g (Ceftriaxone 1000mg + Tazobactam 125mg) once or twice daily
• Severe Infections: May require twice daily dosing
• Duration: Typically 5-14 days depending on infection type and severity
• Dosage adjustments may be necessary for patients with renal impairment

Administration

• For Intravenous (IV) Use:
  1. Reconstitute with 10ml sterile water for injection
  2. Further dilute in 50-100ml of compatible IV fluid (0.9% sodium chloride or 5% dextrose)
  3. Administer by slow IV infusion over 30 minutes
  4. Do not mix with other medications in the same infusion
• For Intramuscular (IM) Use (when IV not possible):
  1. Reconstitute with 3.5ml of 1% lidocaine solution (without epinephrine)
  2. Inject deeply into a large muscle (gluteal muscle or lateral part of the thigh)
  3. Do not inject more than 1g at one injection site

Duration of Treatment

• Continue treatment for at least 48-72 hours after signs and symptoms of infection have disappeared
• Complete the full course of treatment, even if feeling better
• Duration varies by infection type, typically 5-14 days

⏱️ Forget/Missed Doses

Since UPRIDEN CEF T is typically administered in a healthcare setting:

• Notify healthcare provider if scheduled dose is missed
• Healthcare provider will adjust timing of subsequent doses
• Do not double the dose to compensate for missed dose
• For home administration (rare), contact healthcare provider immediately for instructions if a dose is missed

🧬 How this Works

UPRIDEN CEF T combines two complementary mechanisms:

1. Ceftriaxone (1000mg):
   • A third-generation cephalosporin antibiotic
   • Binds to penicillin-binding proteins (PBPs) in bacterial cell walls
   • Disrupts cell wall synthesis by inhibiting peptidoglycan cross-linking
   • Causes bacterial cell wall instability and eventual cell lysis (death)
   • Has broad-spectrum activity against many gram-positive and gram-negative bacteria
   • Long half-life allows for once or twice daily dosing
2. Tazobactam (125mg):
   • A beta-lactamase inhibitor
   • Binds irreversibly to beta-lactamase enzymes produced by resistant bacteria
   • Prevents these enzymes from inactivating ceftriaxone
   • Extends ceftriaxone's spectrum to include beta-lactamase producing strains
   • Has limited antibacterial activity on its own
   • Protects ceftriaxone from degradation, enhancing its efficacy

The combination provides effective treatment against a broader range of bacterial pathogens than ceftriaxone alone, including those that have developed resistance through beta-lactamase production.

Storage Instructions

For Healthcare Providers:

• Store unreconstituted powder at room temperature (20-25°C)
• Protect from light and excessive heat
• Store reconstituted solution between 2-8°C if not used immediately
• IV solution is stable for 24 hours at room temperature or 3 days if refrigerated
• IM reconstituted solution should be used within 1 hour
• Discard any unused portion after the indicated time

For Patients (home administration):

• Follow specific storage instructions provided by your healthcare provider
• Keep all medications out of reach of children

⚠️ Side Effects

Common Side Effects

• Injection site reactions (pain, inflammation, hardness)
• Gastrointestinal disturbances (diarrhea, nausea, vomiting)
• Elevated liver enzymes (transient)
• Changes in white blood cell counts
• Headache
• Dizziness

Less Common Side Effects

• Rash or itching
• Fever
• Elevated blood urea nitrogen (BUN) and creatinine
• Altered taste
• Candidiasis (yeast infection)

🔁 Over Doses

Symptoms of Overdose

• Neuromuscular hyperexcitability
• Seizures
• Encephalopathy
• Severe nausea/vomiting
• Bleeding abnormalities

Action in Case of Overdose

• Seek emergency medical attention immediately
• No specific antidote exists
• Treatment is supportive and symptomatic
• Hemodialysis or peritoneal dialysis are not effective in removing significant amounts of ceftriaxone or tazobactam

Pregnancy and Breastfeeding
